    Northern Hemisphere blocking simulation in current climate models: evaluating progress from the Climate Model Intercomparison Project Phase 5 to 6 and sensitivity to resolution

    Global climate models (GCMs) are known to suffer from biases in the simulation of atmospheric blocking, and this study provides an assessment of how blocking is represented by the latest generation of GCMs. It is evaluated (i) how historical CMIP6 (Climate Model Intercomparison Project Phase 6) simulations perform compared to CMIP5 simulations and (ii) how horizontal model resolution affects the simulation of blocking in the CMIP6-HighResMIP (PRIMAVERA – PRocess-based climate sIMulation: AdVances in high-resolution modelling and European climate Risk Assessment) model ensemble, which is designed to address this type of question. Two blocking indices are used to evaluate the simulated mean blocking frequency and blocking persistence for the Euro-Atlantic and Pacific regions in winter and summer against the corresponding estimates from atmospheric reanalysis data. There is robust evidence that CMIP6 models simulate blocking frequency and persistence better than CMIP5 models in the Atlantic and Pacific and during winter and summer. This improvement is sizeable so that, for example, winter blocking frequency in the median CMIP5 model in a large Euro-Atlantic domain is underestimated by 33 % using the absolute geopotential height (AGP) blocking index, whereas the same number is 18 % for the median CMIP6 model. As for the sensitivity of simulated blocking to resolution, it is found that the resolution increase, from typically 100 to 20 km grid spacing, in most of the PRIMAVERA models, which are not re-tuned at the higher resolutions, benefits the mean blocking frequency in the Atlantic in winter and summer and in the Pacific in summer. Simulated blocking persistence, however, is not seen to improve with resolution. Our results are consistent with previous studies suggesting that resolution is one of a number of interacting factors necessary for an adequate simulation of blocking in GCMs. The improvements reported in this study hold promise for further reductions in blocking biases as model development continues

Further, Action Design Research (ADR) has recently developed as a broad research method that focuses on designing and redesigning IT and IS in organizational contexts. This paper reflects on applying ADR in a complex organizational context in a developing country. It shows that ADR requires additional lens for designing IS in such a complex organizational context. Through conducting ADR, it is seen that an ethnographic framework has potential complementarities for understanding complex contexts thereby enhancing the ADR processes. This paper argues that conducting ADR with an ethnographic approach enhances design of IS and organizational contexts. Finally, this paper aims presents a broader methodological framework, Action Design Ethnographic Research (ADER), for designing artefacts as well as IS. This is illustrated through the case of a land records updating service in Bangladesh

    How do patients and their family members experience the transition from peritoneal dialysis to incentre haemodialysis? A multisite qualitative study in England and Australia.

    BACKGROUND: While numerous studies have explored the patient experience of dialysis or other end-stage kidney disease (ESKD) treatments, few have explored the process of transitioning between dialysis modalities. This study aimed to develop an in-depth understanding of patient and caregiver perceptions and experiences of the transition from peritoneal to haemodialysis (HD) and to identify ways in which transitions can be optimised. METHODS: Fifty-four in-depth, semi-structured interviews were undertaken at six study sites across the West Midlands, UK (n = 23), and Queensland, Australia (n = 31). Thirty-nine participants were patients with ESKD; the remainder were family members. An inductive analytical approach was employed, with findings synthesised across sites to identify themes that transcended country differences. RESULTS: Of the 39 patient transitions, only 4 patients reported a wholly negative transition experience. Three cross-cutting themes identified common transition experiences and areas perceived to make a difference to the treatment transition: resistance to change and fear of HD; transition experience shared with family; and bodily adjustment and sense of self. CONCLUSION: Although each transition is unique to the individual and their circumstances, kidney care services could optimise the process by recognising these patient-led themes and developing strategies that engage with them. Kidney care services should consider ways to keep patients aware of potential future treatment options and present them objectively. There is potential value in integrating expert support before and during treatment transitions to identify and address patient and family concerns

    ReQTL: Identifying correlations between expressed SNVs and gene expression using RNA-sequencing data

The final version is available on open access from OUP via the DOI in this recordBy testing for associations between DNA genotypes and gene expression levels, expression quantitative trait locus (eQTL) analyses have been instrumental in understanding how thousands of single nucleotide variants (SNVs) may affect gene expression. As compared to DNA genotypes, RNA genetic variation represents a phenotypic trait that reflects the actual allele content of the studied system. RNA genetic variation at expressed SNV loci can be estimated using the proportion of alleles bearing the variant nucleotide (variant allele fraction, VAFRNA). VAFRNA is a continuous measure which allows for precise allele quantitation in loci where the RNA alleles do not scale with the genotype count. We describe a method to correlate VAFRNA to gene expression, and assess its ability to identify genetically regulated expression solely from RNA-sequencing (RNA-seq) datasets.We introduce ReQTL, an eQTL modification which substitutes the DNA allele count for the variant allele fraction at expressed SNV loci in the transcriptome (VAFRNA).     A conceptual model for action and design research

    Organizational research has a pattern of special characteristics which make a clear distinction from other research paradigms. When using these approaches – based on Action and Design – the Interpretivist, Constructivist, and Participatory perspectives dominate. They have already proven to have strong foundations, which turn these paradigmatic approaches into effective ways for getting knowledge, doing things, and promoting change within organizational settings. It combines the traditional scientific, engineering, and organization development approaches, depicting how an organization can, simultaneously, solve multidimensional problems and produce actionable knowledge, effective change and useful artifacts. It has been developed using a Design Science Research approach, tested in a major organizational change program (Henriques, 2015; Henriques & ONeill, 2014), and successfully used to teach research methods essentials to Master and DBA students.info:eu-repo/semantics/publishedVersio
